Non-alcoholic fatty liver disease, or NAFLD, is becoming a global health concern. One of its key features is the buildup of fat droplets, which causes stress on the endoplasmic reticulum (ER) and plays a big role in the disease. Scientists created four special fluorescent probes, named COB1 to COB4, that can watch changes in fat droplets (LDs) and the ER inside cells in real-time. These probes were designed with different side chains attached to carbazole groups, while coumarin connected with carbazole acted as precursors.
Out of these probes, COB2 was picked for further study because it had the best ability to resists unwanted interference. Tests showed that COB2 could pinpoint both fat droplets and the ER at the same time, and it could show changes in these organelles under different health conditions. Plus, it could tell the difference between a normal liver and a fatty liver at the tissue level. These findings are a step forward in developing new probes that can target two things at once. They could help in the early diagnosis of NAFLD, which is great news. Keep in mind, though, that more research is needed before these probes can be used in real-life situations.
